From the Eleusinian Mysteries—the most significant esoteric rituals of ancient Greece, centered around the enigmatic kykeon—to the ceremonial use of teōnanācatl, the psilocybin-containing mushrooms of Mesoamerica, ancient cultures harnessed the power of entheogens for spiritual communion, healing, and transcendence. Bridging these ancient traditions and modern science, Dr. Evangelos Dadiotis, a pharmacist and postdoctoral researcher at Northeastern University, Boston, and Romanos Konstantinos Antonopoulos, M.Sc., an agronomist collaborating with the National and Kapodistrian University of Athens and the University of Granada, present groundbreaking insights into the entheogenic potential of psychedelic tryptamines. This presentation delves into the psychoactive constituents of kykeon, with particular emphasis on alkaloids from the ergot fungus (Claviceps purpurea), hypothesized to be the entheogenic agent in this ancient ceremonial beverage. Supported by archaeological evidence and theories by Dr. Albert Hofmann, Dr. Carl Ruck, and Gordon Wasson, this work illuminates the role of ergot-derived compounds in the Eleusinian Mysteries.Additionally, the session explores advancements in psilocybin research, focusing on innovative breeding techniques and biotechnological cultivation methods, including liquid fermentation. Central to this research is the application of quantitative nuclear magnetic resonance (qNMR) spectroscopy, which offers rapid, accurate analysis of tryptamines in different matrices. qNMR delivers transformative potential by enabling direct compound quantitation in under three minutes.By integrating ancient knowledge with cutting-edge scientific methodologies, this research seeks to revive the wisdom of entheogenic rituals while laying the groundwork for the safe and controlled use of psychedelics in modern contexts. This work aspires to serve as the “stomach” of the psychedelic renaissance—digesting ancient traditions and reimagining them in ways that inspire awe and wonder while advancing humanity’s understanding of the therapeutic and spiritual potential of psychedelics.
